Science is the cornerstone of progress. The Science community is working to understand how brain tumors develop in children. This understanding will help to improve outcomes by translating the latest findings/research in brain tumor biology into better treatments for these children.

Working Groups foster collaboration between physicians and scientists as well as leaders of foundations that support medulloblastoma research. The Groups help to refine the design of future clinical trials, creating new treatment options and drug delivery methods, generate a deeper understanding of the mechanisms of metastasis, and develop a broader understanding of late effects and complications of treatment.

Target

Research Questions

New staging systems.

In addition to the staging criteria currently used, newer factors are being examined to help stage medulloblastoma in order to choose treatment. For example, tumors with certain features are being treated as high-risk tumors in some clinical trials. This includes features called anaplastic and changes to certain genes, such as MYC and MYCN.

Molecular testing of the tumor and targeted therapy.

Researchers are looking at laboratory tests for tumor samples to identify specific genes, proteins, and other factors unique to medulloblastoma. It is now known that medulloblastoma is made up of 4 subtypes: WNT, SHH, Group 3, Group 4. Results from these studies may help develop specific treatments for each type of medulloblastoma.

Improved methods of imaging and surgery, radiation & combination therapies.

Areas of research include studies that examine the safety of reducing the doses of radiation therapy for children with a standard-risk tumor by using new approaches to chemotherapy. Other studies in infants and older children with a high-risk tumor focus on new drugs and combinations of radiation therapy and chemotherapy to slow or stop tumor growth. Recent efforts have shown promise.

PNOC

The Medulloblastoma Working Group is a multidisciplinary group of physicians, scientists and parents/advocates committed to improving outcomes for patients with medulloblastoma.

COG

The Children’s Oncology Group partners with research scientists from around the world, develops clinical trials, and offers support to families and patients.

COG priorities

The COG, with more than 10,000 experts worldwide, has nearly 100 active clinical-translational trials open at any given time. These trials include front-line treatment for many types of childhood cancers, studies aimed at determining the underlying biology of these diseases, and trials involving new and emerging treatments, supportive care, and survivorship.

PNOC Priorities

Topics of discussion have ranged from the mechanisms of metastasis and the structure of the blood brain barrier to strategies for immunotherapy (CAR-T cells and RNA vaccines) to the safety and ethics of research biopsies. Going forward we (PNOC) will continue our discussions of topics of interest, with future sessions planned on drug delivery as well as on late effects and complications of therapy.
